Default Using the dot com, is redirect the best thing to do with the .co.uk?

I'm starting an online shop with a friend. We have both the .com and the .co.uk registered.

For branding purposes we are going to use the .com (most uk shops do this too eg firebox, play etc).

Is simply redirecting the .co.uk to the .com the best thing to do? (in my opinion it is!)

Yes 301 is the correct way to go. The other option would be to effectively two identical sites, but then you run into dual content issues with search engines, plus you may confuse your visitors.
